what is B-max?
the maximum binding capacity gives us information about receptor number
What is Kd?
the dissociation constant or the concntration of drug that gives 50% occupancy
What is Kd an index of?
it is the receptrical of affinity low Kd is a higher affinity
What is IC50?
the inhibition constant or the coinctration of ligand needed to displace 50 of the prevoously bound ligand - not the same as Kd
does binding = functional activity?
no
What is efficacy?
the ability of an agonist to cause a response in a cell or tissue.
What is intrinsic efficacy?
the ability of an agonist to activate the receptor not the same as efficacy.
what term do we use to quantify agonist function?
EC50 - effective concentration that gives 50% of the maximal response.
What is potency measured in
EC50
What do we see because of spare receptors?
less than 100% binding we can have 100% response
why do we see spare receptors?
signal transduction pathway is amplified, and the respose is limited by a post receptor event.
Why would we have spare receptors?
allows for functional responses at low-levels of agonist
and the number of receptors can then control potentency.
What is a partial agonist?
an agoinist that cannot elceit the full responce of th endogenous agonist, as they have lowerer intrinsic activity
