If you take agnostic chronically, how does tolerance occur?
tolerance caused by receptor down regulation. Fewer receptors, so more agonists needs to be given to stimulate limited receptors.
If you take antagonists chronically, how does tolerance occur?
tolerance caused by up regulation. So more receptors, more antagonists needs to be given to block the increased receptors
Receptor bonds from weakest to strongest
van der Waals, hydrophobic, hydrogen, ion, covalent
define inotropic receptor
one type of nervous system receptor. also known as ligand gated ion channel receptor. e.g. Gaba receptor
define metabotropic receptor
indirectly linked with channels, like G protein coupled receptors
albumin bind to what drug
acidic drugs
alpha 1 acid glycoprotein and beta globulin binds to what drug
basic drugs
graded dose response
describes response compared to dose of drug. E.g. how much does a vessel constricts with increased norepinephrine dose?
quantal dose response
It describes the % of a certain population responds to a certain dose of drug. E.g. % of people fall asleep given 5mg/kg propofol bolus. LD50 and ED50 are from quantal dose response.
