Principle of wintrobe
Anticoagulated blood
-centrifuged
-optimum,speed radius duration
-RBC-bottom and plasma on top
-ratio of settled cell in% is calculated as hematocrit
Method of wintrobe
-blood in EDTA in tube and mix
-fill with Pasteur pipette to avoid bubbles
-tip of pipete gradually withdrawn from tube by simultaneous expulsion of blood
Points to note in wintrobe
-hemolysis blood is prevented
-upto 10 mark
-centrifuge at 300 rpm at 30 min
-stop after exact time, mark reading
Layers after centrifuge in wintrobe
Lower-packed red cells
Middles-buffy coat (thin greyish)-wBC and platelets
Upper-plasma layer -straw coloured
Colour change in wintrobe
Red color-hemolysis ,faulty technique/wet syringe
