pag 1 microscopy Flashcards
blood smear study mitosis examine lung tissue
magnification of objective lenses used
10x and 40x
stain used
Leishman’s stain
why must you focus stage down
to avoid broken cover slip
how to avoid contamination of slide surface
handle slides by edges
reason for gently shaking sample of blood
to re-suspend the cells
purpose of hypochlorite
clean contaminated equipment
how to smear the blood
2nd slide @ 45’, push backwards until touches drop of blood, then pull towards far end of side
why is it important that the second slide makes the briefest possible contact with the drop of blood
to prevent to much from being collected and far too many cells being spread on the slide
why is the smear left to air dry
so the cells will stick to the slide
why is ethanol poured onto dry smear (for 2min)
to fix and dehydrate the cells
where is the cover slip placed
at end where smear is thinnest
purpose to using x10 objective
to focus the microscope and identify right part of smear
