what are the oris?
pMB1 ori; pUC ori; pSC101 ori; F’ ori; M13 or f1 ori; 2m ori; SV40 ori
purpose of pMB1?
has low copy number, about 15-20 copies per cell (used in pBR322-derived plasmids)
purpose of pUC ori?
a mutated version of pMB1 that removes control of copy number to yield 500-700 copies per cell
purpose of pSC101 ori?
very low copy number, 5 copies/cell
purpose of F’ ori?
single copy number 1-2/cell, also encodes partition genes from conjugative plasmid F’; ensures the plasmid will be divided between the cells during replication
purpose of M13 or f1 ori?
filamentous phage origin to produce ssDNA which can be packaged into virions
purpose of 2micron ori ?
origin from the yeast 2-micron plasmid to allow yeast propagation
purpose of SV40 ori?
simian virus 40 origin to allow propagation in some eukaryotic cell lines
