Non-CYP450 Mediated Reactions Flashcards
What enzyme catalyses alcohol oxidation?
Alcohol dehydrogenase
What enzyme catalyses N and S-Oxidations?
Flavin Monooxygenase - FMO
What enzyme catalyses oxidative deamination?
Monoamine oxidase - MAO
What enzyme catalyses aldehyde oxidations?
Aldehyde oxidase
How does Disulfiram work?
Disulfiram inhibits aldehyde dehydrogenase, resulting in worse hangovers
Used to treat alcohol abuse
Where is Flavin monooxygenase found and what reactions does it mediate?
Found alongside P450s in liver microsomes
Mediates N- and S-oxidation reactions
What co-factor is necessary for FMO?
NADPH
What’re the two types of Monoamine oxidase - MAO? Where are they found?
MAO-A - found in liver, pulmonary vascular endothelial, GI tract and placenta
MAO-B - found in blood platelets
Both are found in neurons and astroglia
What role does MAO play?
Had a vital role in the inactivation of endogenous neurotransmitters
What compounds inhibit MAOs?
Antidepressants and caffeine
What drug is metabolised by MAO?
Sumatriptan - used for treatment of migraine headaches
MAO-A converts tertiary amine to -COOH
Where is aldehyde oxidase located?
Found in cytosolic compartment of tissues in many organisms
What does aldehyde oxidase do?
Catalyses the oxidation of aldehydes into carboxylic acids
Catalyse the hydroxylation of some heterocycles
Can catalyse oxidation of both CYP450 and MAO intermediate products
What does xanthine oxidase do?
Catalyses oxidation of hypoxanthine to xanthine and further oxidation to uric acid
What does a build up of uric acid result in?
Gout
What rare examples does xanthine oxidase catalyse?
XO is known to catalyse xenobiotic metabolism reactions
Give an example of azo reduction
Prontosil is reduced to Sulphanilamide and an inactive by-product
Sulphanilamide is an active bacteriostatic
What enzymes catalyse azo reduction?
Intestinal microbe reductases
What enzymes catalyse nitro reduction?
Intestinal microflora nitroreductases
What enzymes catalyses sulfoxide reduction?
Methionine sulfoxide reductase
What enzyme catalyses the reduction of epoxides?
Epoxide reductase
What enzyme catalyses the reduction of halogenated hydrocarbons?
Cytochrome P2E1
In presence of NADPH
What’re the key hydrolysis enzymes?
Carboxylesterases
butyrylcholinesterases
Alkaline phosphatases
Paraoxanases
Epoxide hydrolase
How is morphine activated?
Through dealkylation
