Benefits and features of NCAs
Analysis of PK viewing the body as one homogenous compartment - no difference in the PK parameters in different tissues within the body.
Useful in studying food and DDI effects on drug PK.
Usually done with oral doses, looking for changes in CL, Vss, and bioavailability.
Simpler to do - fewer plasma samples needed than multiple compartment analysis.
AUC, AUMC, and Kel in NCA
Trapazoidal method is a way to estimate the AUC.
AUMC is the plot of the product of plasma conc and time vs time.
Kel is elimination rate constant. For oral, estimation of the terminal rate constant. Calculated by estimating the slope of log conc-time curve during the terminal phase (after absorption and distribution).
Kel = 0.693/halflife
MRT is the mean time the drug remains in the body after dosing. MRT = AUMC/AUC
