How do you make human like Ab without immunising people with phage display?
Phage has coat protein with PIII that can be fused with human Ab VH and VL gene, then binds and replicates and enriches in bacteria expressing that antigen.
Problem with phage display dreived Ab?
They may not have high affinity for self molecules and haven’t gone through SHM.
An alternative way to create human-like Ab?
WIth transgenic mice expressing human Ab genes and then immunise them- affinity maturation.
