Q

What are two examples of in vitro assays that can be used to show how drugs are metabolised?

A

Ligand binding assay - radio labelled compound is used, wash through filter with membrane bound receptor, measure residual activity
Competitive binding assay - Measure how much radio labelled compound A displaces unlabelled B

Q

What are the advantages and disadvantages for in vitro drug activity methods?

A
Advantages = Cheap, reproducible, high throughput, possible to study single proteins or receptors in isolation, no testing on animals
Disadvantages = Unrealistic, not possible to measure metabolites, limited pharmacokinetic data.

Q

What are the advantages and disadvantages of in vivo drug activity methods?

A

Advantages - allows measurement of effects without fully understanding the mechanism, cam measure drug and active metabolites, can measure pharmacokinetics and is more physiologically relevant than in vitro.
Disadvantages - Using whole animal (ethically questionable and costly), low throughput, can be difficult to determine mechanisms, negative results are difficult to determine/interpret.
