molecular diagnostics Flashcards
What do you need to know in order to run hybridization or PCR?
know the sequence of the pathogen
what is hybridization used for?
detect and quantify target DNA;
works with DNA or RNA;
then hybridized via southern or northern blotting
What is a southern blot?
DNA
northern blot
RNA
western blot
protein or antibody
eastern blot
post-translational modifications
Describe hybridization
you create a flagged primer, if DNA or RNA is present in the sample then it will bind, then you run it through a gel to separate different desities, then transfer to a membrane
describe pcr
Denature DNA in sample, have primers that attach to each DNA strand and then add dNTPs
Taq polymerase
if you have small sample of DNA then you can amplify it
describe quantitative pcr
used to compare two samples’ (find amount of specific genes);
have to use a probe that lights up when certain sequence is reached?
what is rflp?
forensic analysis;
dna fingerprinting based on differences in DNA;
DNA cleaved and run through gel and compare band travel
what is vntr?
Variable number tandem repeats;
PCR, gel, then compare band location
describe ELISA
detect antigen or antibody concentration;
antibodies are at the bed of the well, add the sample and then wash it, add a flag antibody to observe
what is indirect ELISA?
detect amount of antibody in sample
What is Sandwhich ELISA?
detect the amount of antigen in a sample
