MMB Lab Questions Flashcards
Give two examples of aseptic techniques
Flame the working surfaces of equipment to sterilise them
Work near a Bunsen flame to prevent airborne microorganisms from contaminating workspace
Why do microbiologist usually not where gloves, and when should they be worn
Skin is already a good enough barrier to defend against microorganisms so gloves aren’t necessary. Gloves prevent a greater hazard since they can melt if in contact with a Bunsen flame causing burns.
Gloves are necessary if the skin has been breached e.g. cuts or bruises, since microorganisms can now enter
What would you do after spilling a bacterial culture on a bench
Place tissue on spillage
Soak in disinfectant
Dispose of in a biological waste bin
Name two bacteria from hazard group 2
Echeria coli
Bacillus subtilis
Name two identified hazards present in a microbiology lab
Use of Bunsen burners
Exposure to biological hazards
What is the purpose of the streak plate technique
To separate colonies in a mixed culture
Define the term CFU
Colony forming units
Why are agar dishes labelled at the bottom
The kids can be easily mixed up leading to microorganisms being misidentified
Why are afar plates incubated upside down
To prevent the build up of condensation on the lid of the Petri dish which may affect the culture
Why is plate agar cooled to between 45-50C before it is poured into Petri dishes
Bacteria will die in higher temperatures
Would you expect higher bacterial counts from a spread plate or a pour plate culture
Pour plate results in a lower bacterial count since bacteria inside the algae will have less oxygen and therefore grow less
List the different types of culture agar media
Complex media
Defined media
