MIFlowCyt compliance and data display of flow cytometry data in immunological research Flashcards

1
Q

What is the standard for flow? Name a checklist

A

the checklist (one of them) is MIBBI for the standard, MIFlowCyto.

Q

What is the minimum information to warrant reproducibility of an experiment

A

following the MIBBI standard with complementing “checklist” OMIP for replication in different types of experiments like plant cytometry.

Q

what are some requirements for MIFlowcyte compliance? What is the OMIP?

A

Full gating strategy: position of positive and negative controls, essential stats like % of cells in the region/gate/event, axis should include dye and marker (Ag) used, show if the graph is in lin or log, label axis with color and detectors, color coding population with avoiding common color blindness color, to help with all the requirement the OMIP was developed to reduce time to develop similar panels and provide a starting point and for optimization. the OMIP is the “checklist.”

Q

What is the minimum requirements for data display

A

Label everything. If clustering algorithms are used, display fluorescent intensity distribution for each dye/wavelength, color code each population and label the cell type.

Q

What information do you provide for data reduction and analysis tool

A

Use a source code open to the public and include which one used for analysis in the paper. This is for more complex panels. There are many for different types of experiments, more on page 29
