microscopy + light Flashcards
1
Q
simple microscope
A
- light passes through one lens
- e.e. magnifying glass
2
Q
light microscopes
A
- microscope that uses light to illuminate a specimen
- magnification limited to visible light
3
Q
compound optical microscope
A
- uses light + two or more lenses
- objective + ocular lens
4
Q
stereoscopic miscroscope
A
- 3 dimensional view of a sample
- shows colour + detail of a sample → therefore not useful for cell biology
5
Q
transmission electron microscope (TEM)
A
- beam of electrons transmitted through a sample
- 2 dimensional + not colour view
- shows internal ultrastructure of cells
6
Q
scanning electron microscope (SEM)
A
- multiple receptors
- 3 dimensional view
- shows surface features of a sample
- uses scattered light
7
Q
fluorescence microscope
A
- filters allow certain wavelengths of light through and blocks others
- paired with fluorescent dyes
8
Q
fluorescence
A
light of one wavelength is absorbed and light of a different wavelength is emitted
9
Q
emission + excitation
A
emitted light is always of a lower energy + longer wavelength than excited light
