Microscopy Flashcards
1
Q
Resolution
A
How well a microscope distinguishes between two points that are close together
2
Q
Light microscopes
A
Work by passing light through the specimen
3
Q
Electron microscopes
A
Use electrons rather than light.
Higher magnification and resolution - can see smaller things in much more detail
4
Q
Microscope Practical
A
- take a clean slide and use a pipette to our one drop of water in the middle of it - secure specimen in place, then use tweezers to places you specimen on the slide
- add stain if specimen is transparent - makes specimen easier to see
- place cover slip at one end of the specimen, holding it at an angle with a mounted needle and lower it onto the slide. Press it down gently so no air bubbles are trapped. Clip the slide onto the stage.
- select the lowest powered objective lens
- use coarse adjustment knob to move stage up so the slide is just underneath the objective lens, looking down at the eyepiece move the stage downwards.
- adjust the focus with the fine adjustment knob until a clear image. Position a clear ruler on the stage and use it to measure the diameter of the circular area visible
