Microscopy Flashcards
What are the objective lenses
scanning -4x
low-power- 10x
High power- 40x
oil-immersion-100x
What is the magnificantion of all ocular lenses?
10x
Find the total magnification of the lenses;
scanning-4x
low-power- 10x
High-power- 40x
oil-immersion- 100x
scanning= 40x
low-power= 100x
high-power= 400x
oil-immersion= 1000x
Based on this formula, the shorter the wavelength,
the greater the
resolving power of the lense
. Decreasing the wavelength
will not automatically increase the resolving
power of a lens, because
the visible portion of
the electromagnetic spectrum is very narrow and borders on the very short wavelengths found in
the ultraviolet portion of the spectrum.
Resolving power is also dependent on
the refractive index
This is
the bending power of light passing through air
from the glass slide to the objective lens
refractive index
A rule of thumb is that as the magnification of the lens increases, the distance between the
objective lens and slide, called working distance,
decreases, whereas the numerical aperture of the objective lens increases
working distance: The smaller the objective ex 4x
The further away the working distance, the bigger the magnification the smaller the working distance
