A

1.align the eye piece graticule with the stage micrometer, parallel to one another horizontally.
2.count how many divisions on the eye piece graticule correspond to a set of divisions on the stage micrometer
3. Calculate how much one division equals using the stage micrometer and the eyepiece graticule
4.Count how many eye piece graticule divisions your specimen covers.

How do I prepare a temporary slide?

A
  1. Cut an onion between ur fingers
  2. Using forceps add the thin epidermis layer on the slide
  3. Add a drop of water and the cover slip
  4. Add a drop of dye on the other side and filter paper on the other (irrigation technique)
