MICROSCOPY Flashcards
the part of the microscope
that magnifies the image produced by
the microscope’s objective so that it can
be seen by the human eye
ocular lens
to hold the objective and
ocular lenses at the two ends
body tube
Holds the objective lenses &
attaches them to the microscope head.
nosepiece
Supports the tube and connects it
to the base
arm
magnifies the image
objective lens
hold the slides in place
stage
gather wavefronts from the
microscope light source and
concentrate them into a cone of light
that illuminates the specimen with uniform intensity over the entire
viewfield.
condenser
used to vary the
intensity and size of the cone of light
that is projected upward into the slide
iris diaphragm lever
allows for quick
focusing by moving the objective lens or
stage up and down
coarse-focus knob
sharpen the focus
quality of the image
fine focus knob
One
knob controls forward/reverse
movement of the slide. The other knob
controls right/left movement of the slide
Mechanical stage control knob
responsible for shining
light on the object on the slide
light source
It serves as a support for microscopes
base
turns the illuminator off
and on
on/off switch
When
transporting a microscope from one
location to another, always carry it with
both hands. Hold the microscope’s arm
with one hand and support the base with
the other. This prevents it from tilting or
dropping, reducing the risk of damage.
Proper Microscope Carrying
Use the coarse
focus knob to initially bring the sample
into view, but do this gently. Once the
image is roughly focused, switch to the
fine focus knob for precise adjustments.
Avoid excessive force on these knobs as this could potentially damage the
slide or the microscope’s internal
components.
Focusing Technique
Many
microscopes have adjustable light
intensity controls. If you need to change
the brightness of the light source, do so
gradually. Sudden, drastic adjustments
can cause the light to flicker or even
damage the bulb.
changing the light intensity
When
switching between different objective
lenses, always begin by rotating the
nosepiece to the lowest magnification
lens. This provides maximum clearance
between the lens and the slide. Then,
rotate it to the desired objective gently,
without forcing it.
Objective Lens Rotation
When placing a slide
on the microscope’s stage, ensure it is
properly centered and secured using the
stage clips or other securing
mechanisms if available. This prevents
the slide from shifting or falling during
observation, which could lead to
damage or inaccuracies in your
observations.
securing slides
When carrying the microscope, ______________ - one hand
under the base and one on the arm
hold it
securely with two hands
Keep the microscope _________ at all times
upright
Keep microscope lenses clean with ___________. Do not use
paper towels or your clothing.
lens cleaner and lens paper
Use only the ___________ when using
the high-power objective lens.
fine-focus knob
Remove slides from the microscope
before _______.
putting it away
