Microbiology Lab Practical 01 Flashcards
Who is Antone Van Leeuwenhoek
He is the founder of microbiology, invented the microscope.
Who is Robert Koch
He is a German doctor who identified the bacterium that caused tuberculosis. Developed K-streak
K-streak technique
used to get a pure culture of bacteria
Nomenclature
First part: italicized and capitalized (generic name)
Second part: uncapitalized and specie name
Bacillus
Rod shaped bacteria, gram positive, facultative anaerobe
Aerobes
Bacteria that require oxygen to grow
Facultative Anaerobes
can live with or without oxygen
Obligate Anaerobes
can’t live with oxygen
Clostridium
Found in soil and digestive system
Botulinum: food poisoning (toxin: botox)
diffficile: C-diff
Tetani: tetanus
Saprophyte
An organism that feeds on dead or decaying matter
Catalase Test
used to identify organisms that produce the enzyme catalase and breaks down peroxide
Simple Stain
nonspecific, single dye, determines shape, size arrangement. reads with all microbes
Differential Stain
Multiple dyes that help identify microbes, organisms react in different ways
Simple stain basic dyes
positively charged, binds with negative cells ex methylene blue
Simple stain acidic dyes
negatively charges, bind to positive. Help with background
