Microbio Unit 1 Flashcards
Koch’s Postulates
1) Particular microbe has to be present in diseased cases and not in healthy ones.
2) Microbe has to be isolated from diseased case and grown in a pure culture.
3) Disease pathogen can be inoculated into healthy animal and cause the same disease.
4) Same microbe has to be taken from inoculated animal and be grown in culture again
Acellular Infectious Agents
Prions and Viruses
Cellular Microorganisms
Prokaryotes and Eukaryotes
Eukaryotes Examples
Fungi, Protozoa, Humans
Prokaryotes Examples
Bacteria and Archaea
Bacteria and Protozoa are measured in________
Micrometers (um)
Viruses are measured in ________
Nanometers (nm)
Compound Microscope
1) Contains more than 1 magnifying lens
2) x1000 magnification
3) Bright background
3) Resolving power = 0.2(um)
4) Wave length of visible light limits the size of objects that can be seen!!!!!
5) Eye piece is 10x and also has 4 different magnification.
Total Magnification
magnifying power * magnifying power of objective lens
Phase Contrast Microscopes
Used to observe non-living microorganisms
Florescence Microscopes
Contain built in UV light source which glows against dark background
Electron Microscopes do what to all microorganisms
Kill them when imaging
Transmission Electron Microscope
1) Fires electrons through a thin specimen
2) x1000 better than the compound microscope
3) resolving power in 0.2 nm
4) used to see really tiny microbes like rabies and smallpox
Scanning Electron Microscope
1) Electrons are bounced off of the surface of the specimen
2) Resolving power is x100 less than TEM
3) Used to observe outer surfaces of specimens
TEM and SEM are both what kinds of Images?
Black and white images
