Media Flashcards
Media in a gel or semi-solid form

Nutrient agar, trypticase soy agar, nutrient broth
General isolation media/ supportive media
Sheep blood agar (sba), chocolate agar
Nonselective isolation media
Trypticase soy agar, Brucella agar, or beef heart infusion
with 5% sheep blood
Sheep blood agar
Agar is heated or enzyme treated to hemolyzed rbc –> release NAD V factor) and hemin ( X factor)
Chocolate agar
MacConkey agar, Eosin-Methylene blue (emb)
Differential media
Peptone base with lactose; gram-positive organisms
inhibited by crystal violet and bile salts; neutral red as
indicator
MacConkey agar
Peptone base containing lactose; eosin Y and methylene
blue as indicators
Eosin-methylene blue (emb)
Gram negative broth, selenite broth, tetrathionate broth, thioglycollate broth
Enrichment broth
Peptone base broth with glucose and mannitol (primary CHO); sodium
citrate and sodium desoxycholate (bile salt) act as inhibitory
agents
Gram negative broth
Contains sodium hydrogen selenite (toxic for most
Enterobacteriaceae)
Enhance isolation of Salmonella and Shigella
Selelnite broth
Peptone base broth; iodine and potassium iodide, bile
salts, and sodium thiosulfate inhibit gram-positive
organisms and Enterobacteriaceae
Tetrathionate broth
Pancreatic digest of casein, soy broth, and glucose enrich
growth of most microorganisms; includes reducing
agents thioglycolate, cystine, and sodium sulfite;
semisolid medium with a low concentration of agar
reducing oxygen diffusion in the medium; yeast and beef extracts, vit K1, hemin, resazurin (redox indicator)
Thioglycollate broth
Hektoen enteric (he) agar, Salmonella-Shigella (ss) agar, xylose-lysine-deoxycholate (xld) agar
Selective media
Contains lactose, sucrose, salicin, sodium thiosulfate, ferric ammonium citrate, bromthymol blue and acid fuchsin as indicator
Fermenter cho = yellow-orange
Nonfermenter = green or blue
Hydrogen sulfide producing = black ppt (Salmonella)
Hektoen enteric (he) agar
Agar
Supports growth of N. gonorrhea and H. Influenzae
Chcolate agar
Media in liquid form
Broth
Peptone base with lactose, ferric citrate, and sodium
citrate; neutral red as indicator; inhibition of coliforms
by brilliant green and bile salts
Salmonella-Shigella (ss) agar
Colistin-nalidixic acid (cna), modified thayer-martin (mtm)
Antibiotic media
Chocolate base agar, supplement B, vancomycin, colistin, nystatin, trimethoprim lactate Vancomycin = inhibits gram positive Colistin = inhibits gram negative Nystatin = inhibits yeast Trimethoprim = inhibits Proteus
Modified thayer martin (mtm)
Similar to modified thayer martin (mtm) but anisomycin as yeast inhibitor
Martin lewis medium
Support the growth of most nonfastidious bacteria
General isolation media/ supportive media
Contain a nutrient supplement
Nonselective isolation media
provide distinct colonial appearances of microorganisms to aid in their identification
Differential agar
Peptone base, enriched with solution of 2% hemoglobin or IsoVitaleX (BBL) -dextrose, cysteine, vit B12, thiamine, ferric nitrate
Chocolate agar
Supports growth of anaerobes, aerobes (Pseudomonas),
microaerophilic, and fastidious microorganisms
Thioglycollate broth
Isolation and differentiation of lactose fermenting (pink to red)
and non–lactose-fermenting (clear) enteric bacilli (Shigella)
MacConkey Agar
In certain bacteria, exceptional media components must be used to grow
Fastidious bacteria
Turbidity in broth
A least 10^6 bacteria per mL
Type of media
used to isolate gram negative
Differential media
Yeast extract agar with lysine, xylose, lactose, sucrose,
and ferric ammonium citrate; sodium desoxycholate
inhibits gram-positive organisms; phenol red as
indicator
Xylose fermentation
Lysine dacarboxylation
Hydrogen sulfide production
Xylose-lysine-deoxycholate (xld) agar
Type of media
isolation of stool pathogens
Selective media
selective inhibit gram positive bacteria and gram negative coliform bacteria
Selective media
Selective for N. gonorrhoeae and N. meningitidis.
Supports the growth of Francisella and Brucella
spp.
Modified thayer martin (mtm)
Used to inhibit the growth of one organism while enhancing that of another organi
Enrichment broth
Process growing microorganisms in culture
Cultivation
Used most frequently for stool cultures to inhibit normal flora
Enrichment broth
Isolation and differentiation of lactose-fermenting (purple)
and non–lactose-fermenting (clear) enteric bacilli
Eosin-Methylene blue (emb)
Contains sheep blood agar base, colistin (polymyxin e), nalidixic acid
Colistin = disrupt cell membrane of gram negative bacteria
Nalidixic acid = blocks dna replication in gram negative bacteria
Colistin-nalidixic acid (cna)
Yeast extract, agar, charcoal, and salts supplemented with
L-cysteine HCl, ferric pyrophosphate, ACES buffer, and
α-ketoglutarate
buffered charcoal-yeast extract agar (BCYE)
