Lecture 9 Protein Folding III Flashcards
These diseases of AD, Parkinsons and ALS occur, basically when Proteins are misfolded and then they _________?
Aggregate, They are degraded and they hang out and cause B strands and sheets.
1 idea is that misfolding is likely due to what?
inappropriatly high amounts of Protein- protein interaction due to crowding.
The proteins folded state that is most thermodynamically favored is called its ________ state?
native
Energetically speaking, protein Mis folding is Higher or lower in energy than proper native state folding? and why is this so important to the disease. or unfortunate?
Misfolding is lower in energy state and therfore more stable than the Native state. This means, nothing is getting it to break down or be removed from the cell. bad news
Normal Proteins are more what type of secondary structure and less of what type?
More A helix and less B strand. These diseases reviewed today are caused by the conversion of proteins from A helix to B strand. Which makes them insoluable.
The progression of the amyloid plaques is seeding, fibril formation, and deposit/plaque. Which is actually the most destructive/dangerous and why?
the Fibrils are worse, b/c they move and cause havoc, where the plaques are quarantined and just sit. Plaques make for obvious diagnosis upon autopsy.
explain on the log phase is involved in the development of these plaque diseases?
its about build up to a point of no return and one day, one last helix is converted to Bstrand and then the process does this for many of them, and the cell membrane is compromised.
is the folding process explained as being slower and methodical, or rapid and instantaneous
Rapid
metal ions are so important to the processes of our proteins. What percent of our bodies proteins require specific ions to carry out their function?
at least 25%
Name 4 metals of many that may be required for Protein function?
Mg, Zn, Cu, Ca
What are the 3 heavy metals that are toxic to our body, and why are they toxic?
Cd, Hg, Pb, are inhibitors of protein folding
Fibrils form aggregates and wreak havoc on the cell membrane becuase they are not _________ in water?
soluable
These proteins have been discover in the past 20 years to be infectious, What does this mean/
They can be transmitted cell to cell like a virus causing aggregates and more protein misfolding. rememeber unfolding from crowded cell and protein protein interaction.
note made at end on possible TQ test question.
s. Dimutase I and Dimutase II both have what 2 metal ions required?
I Cu, Zn
II Mn, Fe
