helps determine safety of drug
TI = TD50/ED50
dose causes toxicity in 50% of cases/dose produces therapeutic effect in 50% of pop dosed
larger number indictes a safer drug

absorption, distribution, metabolism and excretion
Bioavailability definition
amount of drug circulating in the blood that is available to act on targets
steady state
desired blood drug level maintained over time
physioloical antagonism
Drug B reduces the effect of Drug A
Additive effect
Drug B adds to the effect of drug A
Potentiation
Drug B does not cause an effect on its own but increases the effect of drug A
Abuse/dependence liability
typially contain CNS acivity that produce euphora, hallucinations, or other mood changes
-must screen new drugs for abuse potential
-rats given known addictive drug such as cocaine then are presented with new drug, preferene for new drug indicates stronger abuse potential
interoceptive properties
what the drug “feels “ like in the body
positive interoceptive effects means animal will work for the drug
drug discrimination paradigm
right lever and left lever prouce food. when given saline only left lever produes food, when given reference drug on right lever produces food. For testing animal is given new drug to see if they press right or left lever
