- there is no product at start of kinetic analysis
- during reaction equilibrium position is established where rate of formation of ES= rate of breakdown of ES
- substrate concetration is always greater than enzyme concentration since enzyme can be recycled
- only the initial velocity is measure hence reverse E+P reaction can be ignored
- enzyme is either present as a free enzym or bound to the substrate
