Examples where CRISPR knock-ins could be used:
•Site-directed mutagenesis = changing an amino acid at a specific position
•‘Repairing’ a small deletion in a gene sequence
•Integrating a tag into a gene / region of genome
kinases:
an enzyme that phosphorylates target proteins
different types of tags:
- affinity tag
- fluorescent tag (GFP)
- degron tag
CRISPR application in agriculture:
•Increase crop yield
•Enhance nutrient content
•Disease-resistance
•Drought-resistance
•Streamlined breeding
CRISPR & global warming:
- altering the gut-microbiome of cattle so that the beneficial digestive bacteria create less methane: massive reduction in global greenhouse gas emissions
problem with CRISPR:
guideRNA will be homologous to several regions of the genome - so may cut somewhere other than the desired locus: this is a major concern over using CRISPR therapeutically
