Lecture 1: Microscopy Flashcards
To learn the different parts of the microscope and their function.
Condenser
Collects and concentrates light upward through the object on the stage.
Stage
Platform that holds slides and has a center hole to allow light to pass through and enter the lenses. Adjustment controls allow for movement in horizontal and vertical directions.
Ocular Lenses
Eyepiece lenses through which the object on the stage is viewed. There may be one, usually 2. Provide additional magnification of 5 or 10 times.
Nosepiece
Holds the objective lenses, which are moved and rotated by means of a knurled ring.
Objective Lenses
Usually 3 different lenses; used to magnify the object on the stage.
Iris Diaphragm
Adjusts the amount of light entering the condenser.
Rheostat
Provides the source of light.
Fine and Course Adjustment Knobs
Allows for focusing of the objective lens.
Base
Supports the microscope and contains the light source.
Arm
Provides support between the tube and base; used for carrying the microscope.
Explain how to calculate total magnification.
Total magnification is calculated by multiplying the ocular lens magnification (10x) by the objective lens value (4x, 10x, 40x, 100x).
Explain how to properly carry a microscope.
A microscope must be held with either 2 hands on the arm, or one hand on the arm and the other hand underneath the base.
Explain how to clean the objective lenses.
Put a drop of lens cleaner on lens paper. Then hold it against the lens for a few seconds to dissolve any contaminants. Then lightly wipe the objective with lens paper.
