LECTURE 1 (Intro) Flashcards

1
Q

Characteristics of plasmids

A
  • extrachromosomal piece of DNA (acts independently)
  • normally circular
  • smaller than chromosome
  • versatile (used for cloning)
  • antibiotic resistance

Q

What is a phagemid?

A

Phagemid: plasmid with a ori from a bacteriophage that allow ss replication and packaging into the phage. (it also has a ds ori needed for ds replication)

Q

What is a cosmid and a cosa site?

A

Plasmid with a cos site from a bacteriophage. The cos site is a sequence essential for packaging.

Q

What is a fosmid?

A

A cosmid based on a F’ plasmid. it has a sequence that allows it to integrate into the bacterial chromosome via homologous recombination.
