Lecture 1 Flashcards

1
Q

Pharmacology

A

The study of the beneficial effects of chemical
agents on biological systems. From the Greek pharmakos

A

The study of the adverse effects of chemical agents
on biological systems. From the Greek toxikos (τοξικός) for

Q

Subdivisions of Toxicology

A
  1. Analytical:
-Clinical: Measuring things, such as in hospitals (urine, blood…etc)
-Forensic: Similar to clinical except that there is a legal aspect to it.
-Biomonitoring: Monitor of biological pollutants in the human body. (Drug Testing)
-Therapeutic Drug Monitoring: Monitoring of drugs for treatments.
  2. Drug Development:
-Pre clinical testing: Animal testing or in vitro.
    -Clinical trials: Human testing.

Q

What is meant when Specificity of response is used?

A

Specificity is targeted to one site. (Like heart medication)

Q

Who said “All substances are poisons. There is none that is not
a poison. The right dose differentiates poison from a
remedy”?

16
Q

Borgias

A

Pope Alexander VI

17
Q

Catherine de Medici

A

Systematic study of the effects of poisons on
the sick and poor to make sure the “correct” formulation was
delivered to her customers.

18
Q

Percival Potts

A

Discovered that chemicals can cause cancer. Scrotal cancer from chimney soot exposure.

19
Q

Mathieu Orfila

A

Forensic/analytical toxicology. Considered to be the first toxicologist.

20
Q

Paul Ehrlich

A

Mechanistic toxicology and phramacology, “magic bullet” concept. (Drug is designed to hit one target).

21
Q

Alice Hamilton

A

Occupational toxicology

22
Q

What did Percival Potts discover?

A

Young poor boys (Age 4-7) that would clean chimneys would after time developed scrotal cancer due to the exposure to chemicals and poor hygiene conditions.

23
Q

~400 BC: Hippocrates

A

discovers pain relief treatments with
powder made from the bark and leaves of the willow tree
(Salix sp.) (Analgesic)

24
Q

1897: Felix Hoffman

A

a chemist at Bayer, synthesized
acetylsalicylic acid and tested it on his arthritis-suffering
father. (aspirin was created).

25
Q

Johann Pagenstecher

A

1830: Swiss pharmacist isolated salicin
as the active ingredient in Meadowsweet

26
Q

Bayer receives patent for acetylsalicylic acid (Aspirin).

A

1899

27
Q

Aspirin sales?

A

Sales today exceed 50 billion pills per year.

28
Q

What is the “horse named jim” incident?

A

– Tetanus contaminated horse serum was used to produce
a diphtheria antitoxin.
– Contaminated vaccine.
– Caused the deaths of 13 children in St. Louis, MO.

29
Q

What did the “horse named jim” incident lead to?

A

The 1906 Pure Food and Drugs Act

30
Q

The 1906 Pure Food and Drugs Act

A

-Food or drug label could not be false or misleading, but no requirement
for safety or efficacy testing.
-Levels of eleven dangerous ingredients, including alcohol, heroin, and
cocaine, had to be listed

31
Q

Elixir of Sulfanilamide

A

– The first antibiotic “wonder drug”.
– In one batch, diethylene glycol (Antifreeze) substituted for
propylene glycol (Used as a way to dissolve drugs).
– Proved deadly; 100+ die after taking
reformulated product (1937)
– Caused acute kidney failure

32
Q

What did the incident with the Elixir of Sulfanilamide lead to?

A

Federal FD&C Act of 1938

33
Q

Federal FD&C Act of 1938

A

Mandated pre-market safety approval of all new drugs, however,
drugs did not have to be proven effective.
Pharmaceutical companies must submit “New Drug Application”
(NDA) to FDA prior to marketing.

34
Q

Thalidomide

A

Used for morning sickness in Europe, caused severe birth defects (Not necessarily lethal).

35
Q

Frances Kelsey

A

FDA reviewer who had concerns of safety about Thalidomide. Drug was not approved for sale in the US.
Lead to the Kefauver-Harris Drug Amendments of 1962

36
Q

Kefauver-Harris Drug Amendments of 1962

A

Mandated both pre-market safety and effectiveness
Regulated prescription drug advertising. (side effects and contraindications).
FDA given right to withdraw drugs from market
