Lecture 1 Flashcards
Pharmacology
The study of the beneficial effects of chemical
agents on biological systems. From the Greek pharmakos
(φάρμακον) for “medicine” and logos (λόγος) for “reason” or
“knowledge”.
Toxicology
The study of the adverse effects of chemical agents
on biological systems. From the Greek toxikos (τοξικός) for
“poisonous” and logos (λόγος) for “reason” or “knowledge”.
Subdivisions of Toxicology
- Analytical:
-Clinical: Measuring things, such as in hospitals (urine, blood…etc)
-Forensic: Similar to clinical except that there is a legal aspect to it.
-Biomonitoring: Monitor of biological pollutants in the human body. (Drug Testing)
-Therapeutic Drug Monitoring: Monitoring of drugs for treatments. - Drug Development:
-Pre clinical testing: Animal testing or in vitro.
-Clinical trials: Human testing.
What did early humans use as natural toxins for?
– Animal venoms.
– Toxic metals.
– Plant extracts.
– Hunting.
– Warfare.
– Assassination.
What is the Ebers Papyrus?
It was a scroll found in Egypt in the 1870s containing 700 drugs in 900 formulas. Contains
indications, dosages, and appropriate spells and incantations.
Hippocrates
Early physician, defined “effective dosages” of toxins and described bioavailability (How well a drug is absorbed).
Socrates
Hemlock poisoning. (Greeks used it as an execution method).
Dioscorides
(Materia Medica; plant, animal, and mineral toxins).
– Discovery of Belladonna (scopolamine).
– Discovery of Foxglove (digitalis).
Avicenna (ibn Sina)
First to suggest clinical trials.
Paracelsus
German physician/alchemist.
– Defined the “toxicon”.
– Experimentation and observation.
– Therapeutic vs. toxic dose.
– Specificity of response
What is meant when Specificity of response is used?
Specificity is targeted to one site. (Like heart medication)
Who said “All substances are poisons. There is none that is not
a poison. The right dose differentiates poison from a
remedy”?
Paracelsus
Who is the “father of toxicology”?
Paracelsus
Lady Toffana
“Queen of Poison”; arsenic cosmetics; established
dose and time relationship.
Hieronyma Spara
“Italian Widows Club”; would sell arsenic for gold.
