Lab Practical I - Microscope Flashcards
1
Q
How do you calculate field of view?
A
- Calculate Total Magnification
- Divide the Total Magnification by the field number
Recalculate if the lenses or microscope are changed.
Also note that this measurement is in millimeters

Q
How to manually calculate the Diameter of the Field of view for a parfocal microscope?
A
- Move the mechanical stage as far as possible to the back of the stage
- Obtain a metric ruler and place it on the stage
- Set the microscope to scanning size
- Alight one of the mm markings so that it is just barely visible on the left hand side of the field of view
- Measure the diameter of field of view by counting the number of spaces (not lines) visible
- Convert mm to micra by multiplying the # millimenters X 1000
- If needed, multiply the DFV in micra by the scanning objective lens magnification. This is a constant value (parfocal lenses only).
