Lab Practical I - Microscope Flashcards

1
Q

How do you calculate field of view?

A
  1. Calculate Total Magnification
  2. Divide the Total Magnification by the field number

Recalculate if the lenses or microscope are changed.

Also note that this measurement is in millimeters

Q

How to manually calculate the Diameter of the Field of view for a parfocal microscope?

A
  1. Move the mechanical stage as far as possible to the back of the stage
  2. Obtain a metric ruler and place it on the stage
  3. Set the microscope to scanning size
  4. Alight one of the mm markings so that it is just barely visible on the left hand side of the field of view
  5. Measure the diameter of field of view by counting the number of spaces (not lines) visible
  6. Convert mm to micra by multiplying the # millimenters X 1000
  7. If needed, multiply the DFV in micra by the scanning objective lens magnification. This is a constant value (parfocal lenses only).
