Lab lecture microscopic exam pt 2 Flashcards
1
Q
Sternheimer-Malbin stain
A
- Most common stain for sediment
- General purpose stain
- Crystal violet and safranin O
- ID WBC, epithelial cells, and casts
2
Q
Toluidine blue stain
A
- 0.5% solution used to enhance nuclear detail of WBCs and RTE cells
3
Q
Oil Red O and Sudan III lipid stains
A
- Stain triglycerides, neutral fats, but not cholesterol
- Appear as free fat droplets, cells containing lipids, or casts
4
Q
What colors do triglycerides, neutral, fats, and cholesterol stain in lipid stains?
A
- Triglycerides and neutral fats stain orange-red
- Cholesterol does not stain, so use polarizer
- All 3 lipids usually appear together in sediment
5
Q
Gram stain
A
Used to ID bacterial casts in UA
6
Q
Hansel stain
A
- ID eosinophils (stain reddish)
- Methylene blue and eosin
7
Q
Prussian blue stain
A
- Stain hemosiderin granules (blue)
- Unstained hemosiderin granules are yellow-brown (seen in RTE cells and casts post-hemoglobinuria)
8
Q
Two ways to clear up amorphous crystals
A
- 2% acetic acid
- 2% sodium hydroxide
9
Q
How does 2% acetic acid affect elements in the sediment?
A
- Enhance nucelar detail of WBC but lyses RBCs
- Yeast and bacteria unaffected
10
Q
How does 3% NaOH affect sediment elements?
A
Destroys cells and casts
