What is the role of agar?
Agar acts as a solidifying agent (melts at 100c and solidifies at 44C)
Slective
permits growth of desired organisms and selects against undesired organisms.
Differential
permits growth of many different types of organisms but allows you to differentiate them based on their appearance.
Enrichment
contains nutrients that enhance the growth of a desired organism.
General media
supports the growth of most bacteria.
MacConkey’s Agar (MAC)
Selective- inhibit growth of Gram-positive bacteria (selects gram-negative)
Differentiates- between lactose fermenters (pink colonies) and non-fermenters (no color change)
Mannitol Salt agar (MSA)
Selective- selects for staphylococcus species (high salt concentration)
Differential (mannitol)- differentiates between mannitol fermenters (yellow colonies) and non-fermenters (no color change)
Phenol red (pink-basic/yellow-acidic)
Phenylethyl alcohol agar (PEA)
Selective- selects for gram-positive (phenylethyl alcohol)
Eosin methylene blue agar (EMB)
Selective-selects for gram-negative.
Differential- differentiates between strong lactose fermenters (green metallic colonies), weak lactose fermenters (pink colonies) and non-fermenters (no color change)
Enrichment (lactose/sucrose)-enhances growth of fecal coliforms.
Blood agar
Enrichment (Blood)- enhances growth of fastidious bacteria (pathogens).
Differentiates- differentiates based on type of hemolysis. Alpha-hemolysis (partial destruction of RBCs, greenish color may develop around colonies), Beta-hemolysis (complete destruction of RBCs, zone of clearance surrounds colony) and y-hemolysis (non-hemolytic/no change)
