- ribbon not stretched out enough in waterbath
- folding within tissue due to excessive compression

Describe an ideal Hematoxylin & Eosin stain
A
- dark purple nuclei
- nuclear detail should be seen
- shades of pink: cytoplasm, muscle, collagen, RBCs

Describe an ideal Periodic Acid Schiff stain
A
- identifies tissue components with large amounts of glycogen (ie. basement membrane)
- aldehydes generated through periodic acid oxidation stains bright magenta with the Schiff’s reagent
- counterstain (hematoxylin) stains background a pale purple

A
- stains collagen green-blue (alanine blue) and muscle fibres red (fuchsin red)
