LAB 5 Quality Evaluation Of Stained Slides Flashcards
1
Q
What causes wrinkles in stained slides ?
A
- ribbon not stretched out enough in waterbath
- folding within tissue due to excessive compression
2
Q
What causes chatters in stained slides ?
A
- Excessive vibration; knife skips sections of tissue
- Incorrect clearance angle
- Cutting too fast
- Tissue too hard
3
Q
What causes scoring in stained slides ?
A
- Nick in knife edge
- Hard particles in tissue
4
Q
What causes stained slides that are not deep enough/ too deep ?
A
Wrong micron setting
5
Q
What causes “parched earth” in stained slides ?
A
- waterbath too hot
- prolonged waterbath exposure
6
Q
What causes scrapes/ damage on stained slides ?
A
- aggressive cutting
- incomplete impregnation of tissue
7
Q
What causes contamination in stained slides ?
A
- waterbath has extra tissue (dirty)
8
Q
Describe an ideal Hematoxylin & Eosin stain
A
- dark purple nuclei
- nuclear detail should be seen
- shades of pink: cytoplasm, muscle, collagen, RBCs
9
Q
Describe an ideal Periodic Acid Schiff stain
A
- identifies tissue components with large amounts of glycogen (ie. basement membrane)
- aldehydes generated through periodic acid oxidation stains bright magenta with the Schiff’s reagent
- counterstain (hematoxylin) stains background a pale purple
10
Q
Describe an ideal Masson Trichrome stain
A
- stains collagen green-blue (alanine blue) and muscle fibres red (fuchsin red)
- cytoplasm stains pink
