L14 Cellular models of neurotoxicity Flashcards
Aims: To discuss examples of cellular models of neurotoxicity due to oxygen deprivation, glutamate and metamphetamine exposure. Outcomes: Familiar with several examples of cellular models Develop the ability to design a cellular model for the study of specific neurotoxins
What effect did oxygen deprivation have on dendrites?
Caused varicosity formation.
Distal swelling at short exposure times, and extensive beading and dendrite loss after long exposure.
What fluorescent membrane tracker dye allows visualisation of dendrites?
Dil?
How are variscosity formations inhibited in oxygen deprived neurones
NMDA antagonist MK-801
What does variscosity formation mean for the neurone?
Preceeds death
What is LHD?
LDH release into media during membrane damage. Is used as an end point measurement.
How can the exictotoxic effects of glutamate be blocked?
NMDA receptor antagonists
What does adding polyphenol chlorogenic acid (CGA) do for neuroprotection?
Protects against glutatmate induced cell death in neurons.
How does CGA work to reduce toxicity?
Inhibits glutamate-induced release of intracellular Ca2+
What role does metamphetamine play in neurotoxicity?
Induces apoptotic cell death in vitro.
Increases expression of COX-1.
Induces protein aggregation.
What role does vitamin C play in metamphetamine toxicity?
Elicits protective effect.
Anti-oxidant. METH induces ROS.
