Competetive Enzyme Inhibitor Drugs
-Competes with substrate by non-covalently binding at active site
-Unchanged Vmax -Higher Km (lower affinity)
Statins: Lowers cholesterol
Ibuprofen: NSAID inhibits cycloxygenase -> No prostoglandin
Captopril: Lowers BP
Active Site-Directed (Transitiion-State) Drugs
-Bind to active site with covalent linkage
Organophosphates: Neurotoxin, inhibits acetylcholinesterase -> no ACh breakdown
Cyclooxygenase: NSAID, inhibits cycloxygenase -> No prostoglandin
Enzyme-Activated (Suicide) Drugs
-Reacts with enzyme and gets altered, making it bind very tightly to enzyme
Penicillin: Antibiotic, inhibits cross-linking in bacterial wall
Allopurinol: Gout Tx, inhibits uric acid formation
