Explain first generation of sequencing: Name, procedure and output

A

Sanger:

  • fragment DNA
  • clone it into plasmids and amplify
  • add DNA polymerase, a primer and tagged nts (+ some ddNTPs to terminate chain)
  • run capillary/gel and read code
