Innate immunity Flashcards
toxins often target
immune cells
some toxins aren’t directly toxic… instead they
induce leukocytes –> produce inflammatory mediators = toxic
toxicity of endotoxin
- not directly toxic
- induces body to produce toxic factors
toxic component of endotoxin
LPS, specifically Lipid A region
endotoxin shock
- Gram negative bacteria
- septic shock
- decreased BP and CO
- ARDS, DIC
Diseseminated Intravascular Coagulation (DIC)
- indicative of septic shock (endotoxin)
- too much clotting –> not enough clotting factors left over –> hemmorhage
ARDS
- acute resp distress syndrome
- gram negative insult to lungs, septic shock
- endotoxin
Pyrogen free
no endotoxin
endogenous mediators of response to LPS
- IL-1
- TNF-a
- produced by LPS-stimulated macrophages
TLR 4
- recognizes LPS
- in plasma membrane of macrophages, dendritic cells, mast cells, eosinophils
IL-1 and TNF-a in liver
complement, opsonization
IL-1 and TNF-a in bone marrow epithelium
neutrophil mobilization –> phagocytosis
IL-1 and TNF-a in hypothalamus
increased body temp –> decreased virus/bacteria replication
IL-1 and TNF-a in fat
increased body temp –> decreased virus/bacteria replication
TLR4 pathway once LPS is bound
NFkB get activated, activates txn of cytokines
