INFILTRATION/ IMPRERGNATION Flashcards
Involves removal of clearing agent from tissues
INFILTRATION
Purpose/s: OF IMPREGNATION/ INFIL
- To remove clearing agent from tissues
- to fill up cavities & tissue spaces for tissues
to have firm consistency in order to facilitate
easy cutting
Melting point of paraffin wax commonly used
56°C
Most common method that uses heat
application
● Not suited for fatty tissues
Paraffin Wax Infiltration
Uses an oven
➔ Temperature range of paraffin oven when in
used: 55-60°C
➔ Approximate temperature of paraffin oven
when in used: 2-5°C higher than the wax
melting point\
➔ Requires 4 changes of wax
Manual
ses an autotechnicon
➔ Requires 3-4 changes of wax
AUTOMATIC
Infiltration under negative atmospheric
pressure inside the oven
➔ Tissue processing time is reduced
➔ Facilitates removal of transition solvents
➔ Prolongs life of wax by reducing solvent
contamination
Vacuum
Substitutes for Paraffin Wax
Paraplast
Bioloid
Carbowax
Ester Wax
Method Recommended for specimens with
large and hollow cavities that tends to collapse
● Disadvantage: Process is slow
Celloidin/Collodion Infiltration
Celloidin infiltration method recommended
forwhole eye specimens
Dry Celloidin
Celloidin infiltration method for bones,
brain & teeth specimens
➔ Will require 70-80% alcohol to store blocks
Wet Celloidin
Thin
2-14%
5-7 days
Medium
4-6%
5-7 day
Thick
8-12% (very high)
3-5 days
Method for enzyme & histochemical studies
● Not usually done in the lab
● Recommended only if dehydration is to be
avoided
Gelatin Infiltration
THICKNESS OF GELATIN
Will require tissues not to be more
than 2-3mm thick
Serves to prevent the growth of
molds
Use of Pheno
Under celloidin infiltration
● Soluble in equal concentration of Alcohol and EtheR
Nitrocellulose Method
Plasticizers to prevent cracking tissues when
infiltrated using LVN (Low Viscosity
Nitrocellulose
Oleum Ricini & Castor Oil
Can be used in high concentrations
● More preferred in both embedding and
infiltration method
● Harder tissue blocks are produced
● Allows cutting of thrinner sections
LVN
