a protein phosphatase that allows NFAT to enter nucleus - causing transcription of IL-2 genes
Sirolimus MOA
mTOR inhibitor - binds FKBP, which prevents response to IL-2
Sirolimus Toxicity
NOT Nephrotoxic
Anemia, thrombocytopenia, insulin resistance, hyperlipidemia
Daclizumab, Basilixumab MOA
Blocks the IL-2 Receptor
Azothioprine MOA
Antimetabolite precursor of 6-Mercaptopurine, blocks nucleotide synthesis to prevent lymphocyte production
Azothioprine Toxicity
Leukopenia, Thrombocytopenia, Anemia
Toxicity is increased by Allopurinol, which prevent Xanthine Oxidase from degrading 6-MP.
Mycophenolate Mofetil MOA
Inhibits Inosine Monophosphate Dehydrogenase
Glucocorticoids MOA
Inhibits NF-kB
