immune pharm Flashcards
Cyclosprine
MOA
Calcineurin inhibitor;
binds to Cyclophilin
blocks T cells activation by preventing IL-2 transcription
cyclosporine
main side effect
nephrotoxicity
other: viral infection, lymphomas. HTN (vasoconstriction of eff/aff artioles in kidney)
gingival hyperplasia, hirtuisim
Tacrolimus
pimecrolimus
tacrolimus - systemically given for transplant rejection
both used for topically for eczema
Tacrolimus (FK506) and pimcrolimus
MOA
CALCINEURIN INHIBITOR
bind to FK506 binding protein (FKBP) –>
Blocks T cell activation by inhbiting production (transcription) of IL-2
tacrolimus (FK506) and Pimecrolimus
main sideeffects
both nephrotoxicity( bc calcineurin inhibitor)
tacrolimus - Neurotoxicty!!! (HA, parethesias)
Sirolimus (Rapamycin)
mTOR inhibitor
binds FK binding protein -12 (FKBP-12) –> inhibits mTOR (mammalian target of rapamycin) –>Blocks T cell activationand B-celll differentiation by preventing response to IL-2
Main side effect of sirolimus (rapamycin)
not nephrotoxic!!
Mycophenolate mofetil
moa?
used for lupus
reversibly inhibits inosine monophosphate (IMP) dehydrogenase –> inhibits purine GUANINE synthesis of B and T cells (prevents lymphocyte proliferation)
Thalidomide
main sided effect if given to pregnant mom
(teratogenic)
Phocomelia
(hyperplasia fused fingers/hands)
Thalidomide
MOA
Suppresses TNF-a production,
increases NK cells and IL-2
the suppresson of TNF a tx erythema nodosum leprosum (hansen disease)
inc nk cells –> tx multiple myeloma
infliximab
adalimumab
MOA?
Etanercept (decoy receptor)
moa
TNF-a blockers
etanercept (decoy receptor) it mimics TNF-a receptor and binds it all up
what drug binds to CD 20
used to treat
B cell non hodgkin lymphoma
CLL
RA, ITP
Rituximab
what drug targets IgE
txs refractory allergic asthma
prevents IgE binding to FceRI
omalizumab
digoxin immune Fab
txs?
digoxin overdose
it targets digoxin
what drug is an antibody toward
platelet glycoproteins IIb/IIIa
Abciximab
” IIb * IIIa = absiximab”
used in pts that have mi, unstable angina
