Prick thumb by lancet
Close one end of tube by wax
Centrifuge for 5 minutes
Read pcv against scale where rbcs at level of 0 while plasma at other level
Principle
Cells are separated from plasma by specific gravity
Rbcs 1090,
Wbcs and platelets or puffy coat 1060,
Plasma 1030 so bottom layer forming then puffy coat middle then rbcs at top coat
Values
Males are 46% +- 5%
Females are 42% +- 5%
Variations
High pcv due to polycythemia
Dehydration by vomiting, diarrhea and profuse sweating
Venous blood
Low pcv due to anemia
Over hydration or pregnancy
Arterial blood
Why hematocrit value higher in venous blood
Due to chloride shift
Pcv in high altitudes
Hypoxia cause erythropoietin formation from kidney
So increased rbcs production so higher pcv
Pcv Microcytic hypochromic anemia
