Gram staining is most consistent on cells for how long?
A
Less than 24 hours

Why is gram staining only consistent for a short period of time?
A
Because young cells are less likely to have cell wall damage

What are the steps for obtaining a smear?
A
- Draw 2 dime sized circles on back of slide and label each circle
- Prepare the smear
- Flame the loop until red hot and let cool
- Pass tube through flame and use loop to obtain broth culture.
- Pass loop through flame again before recapping tube.
- Spread culture on 1st circle of slide. Repeat process to have 2-3 cultures on slide.
- Let smear dry for 30 minutes
