Pyruvate kinase deficiency
Autosomal recessive
Enzyme to have low affinity for phosphoenolpyruvate –> thermally unstable + lower inhibition constant for ATP
Chronic haemolytic anaemia (red blood cell destruction)
Untreated Type I Diabetes mellitus
Net mobilisation of glycogen –> gluconeogenesis –> exceedingly high blood glucose conc.
Maternal hypoglycaemia
In pregnancy, when foetal glucose demand is high
Fructose intolerance
Affecting glucokinase (liver) Absence of end-product inhibition --> large quantities of F-6-P generated --> can't be metabolised further --> liver ATP levels drop --> compromising hepatocyte cellular function
Lactic acid build up
Lowers pH
Especially likely when blood flow inadequate : extreme exercise, angina pectoris attack
