Type of genetic variation in phage
-Conditional mutant: the mutant phenotype is express at a particular condition know as restrictive condition . WT is seen in permissible condition. include temperature sensitive and host range
-unconditional mutant: the mutant is alway express (include plaque morphology mutant)
Recombination process in phage
-coninfection of host with high multiplicity of infection (MOI = 5)
-create a partial diploid stage and permit recombination between two phage dna
-recombination can occur early or late, more than once leading to different classes
–collect the progeny phage to score for recombination (MOI <1)
calculate RF in phage
when the phage parental are unable to grow in restrictive condition
RF= 2xWT/ total
when all type can be distinguish physically
RF= total recombinant class/ total
implication of rf in phage
-if two genes are close, RF is lower
-by looking at assay for low recombinant, a fine genetic map can be constructed
