How do you make cDNA?
First, isolate mRNA from cells, then mix with free DNA nucleotides and reverse transcriptase, the reverse transcriptase uses mRNA as a template to synthesis new strands of cDNA
Why do we need cDNA?
to form DNA fragments containing the target gene to transfer a gene from one organism to another
What are the three ways of producing DNA fragments?
Using reverse transcriptase
Using restriction endonuclease enzymes
Using a gene machine
Describe how we use reverse transcriptase to create DNA fragments:
Because most cells only contain 2 copies of a specific gene, they are difficult to obtain, but cells that produce the proteins coded for by the target gene will contain many mRNA molecules complementary to the gene
We use reverse transcriptase to create complementary DNA from the mRNA molecules.
