Temperature
Increasing the temperature to the optimum will increase the kinetic energy meaning more enzyme-substrate molecules will be formed. This means the rate of reaction will increase.
Increasing the temperature above the optimum will increase the kinetic energy so the active site shape changes so it no longer complimentary to the substrate. this means less/no enzyme-substrate molecules will be formed, slowing down the rate of reaction.
PH
Increasing/decreasing above/below the optimum PH will cause the active site shape to change so that it is no longer complimentary to the substrate. This means that less/no enzyme-substrate molecules are formed, slowing down the rate of reaction
