Exploitaion of the actin cytoskeleton by Listeria Flashcards
What is alpha-actinin?
a cross linking protein
What does Listeria express?
ActA at pole of bacterium
What happens when ActA binds actin in the host cytoplasm?
it is only sufficient to allow limited motility
What is required for full motility?
VASP binding (proline-rich region of protein)
What does VASP do?
recruits Arp complexes –> increases comet tail formation
binds profilin –> good ATP-containing G actin molecule supply for polymerisation
How does Listeria bind to the host cell?
Outside –> via E-cadherin
What does Listeria binding trigger?
Normal endocytosis/phagocytosis mechanisms
How does Listeria access the cytoplasm?
once bacteria inside phagolysosome –> produces lysin proteins –> breaks down organelle membrane –> Listeria accesses cytoplasm
How does Listeria move through the cell + between cells?
intracellular motility
