Experiment: investigate the effect of heat denaturation on enzyme (catalase) Flashcards
1
Q
What enzyme is used?
A
Catalase
2
Q
What substrate is used?
A
Hydrogen peroxide

Q
List the necessary steps for this experiment
A
- Blend celery and water for 4 seconds and filter using coffee paper.
- Pour filtered solution into a graduated cylinder and place in a water bath of 25 degrees C.
- Place pH buffer solution 9 in the graduated cylinder.
- Add one drop of washing-up liquid to the graduated cylinder.
- Add some hydrogen peroxide to a smaller graduated cylinder. Place in the water bath.
- Leave both in a water bath until they have reached 25 degrees C.
- Remove and pour hydrogen peroxide into the graduated cylinder containing the blended celery.
- Note and record the volume at the top of the foam after 2 minutes.
- Repeat the past steps using boiled catalase solution instead of catalase solution as it denatures the enzyme.

Q
How much foam will be produced with catalase solution and with boiled catalase solution?
A
None will be produced with boiled celery as the enzyme has been denatured while foam will be formed with normal celery
