Purpose
To compare the action of catalase to a non-protein catalyst under different conditions
Materials (5)
3% hydrogen peroxide manganese dioxide fresh or frozen liver potato ice
Equipment (6)
fine clean sand stirring rod bunsen burner or hot plate 250 mL beaker test tubes scalpel
How many test tubes are needed? (max-min)
4-11 test tubes
Procedure (Headings) (A-E)

Procedure: A: Catalytic Reactions
Add 2mL hydrogen peroxide to 2 test tubes
Place 0.1g of sand in one test tube
Add 0.1g of manganese dioxide in the second test tube
Observe & Record reaction rates
Procedure: B: The effect of an enzyme
Add 2mL of hydrogen peroxide to 2 clean test tubes
Place a small piece of liver in one test tube
Place a small piece of potato in the other test tube
Observe, Record and Compare (w/ A)
Procedure C: Re-using an enzyme
Divide liquid portion of the previous tube in B w/ liver into 2 test tubes
Cut the liver from B into 2 equal parts & add to the 2 test tubes
Add another piece of liver to one of the test tubes
Add 1mL of hydrogen peroxide to other test tube
Observe & Record
Procedure D: Effect of particle size
Place a small piece of liver in one test tube
Place a small piece of potato in another test tube
Add a pinch of sand to each tube
Crush these materials (out of the test tubes) with a stirring rod
Add 2mL of hydrogen peroxide to each test tube
Observe, Record and Compare with part B
Procedure E: Effect of Temperature
Place a small piece of liver in a test tube & heat for 5 min in boiling water bath
Add 2mL hydrogen peroxide to the boiled liver
Record results
Place a small piece of liver into each of two test tubes
Place one test tube in 37 degree water for 5 min
Place one test tube in an ice water bath fro 5 min
Remove both test tubes from water
Add 2mL hydrogen peroxide into both tubes
Record the rates of reaction
