enzyme controlled reactions Flashcards

1
Q

what is the experiment to see how fast the product of a reaction appears, describe it

A

set up boiling tubes with the same amount of sodium peroxide add equal volumes of a suitable buffer solution
set up the apparatus
put each boiling tube in a water bath at different temperatures with another tube containing catalyse wait 5 minutes so the enzyme gets to temperature
use a pipette to add the same volume and concentration of catalyse to each boiling tube and quickly attach the bung and delivery tube
record how much oxygen is produced in the first minute , use a stopwatch to measure
repeat the experiment three times and use results to find the mean volume of oxygen produced

Q

what is the experiment to see how fast the substrate is broken down, describe it

A

put a drop of iodine in potassium iodide solution into each spotting tile
mix together a know concentration and volume of amylase and starch in a test tube
use a dropping pipette to put a drop of this mixture into one of the wells containing the iodine solution at regular intervals
observe the resulting colour
you can see how fast amylase works by recording how long it takes for the iodine solution to no longer turn black/blue when the starch/amylase mixture is added
repeat experiment using different concentrations of amylase
repeat 3 times and find a mean time taken
