What is an endocrine drug?
Endocrine drugs are agents directed to a malfunctioning endocrine path.
What is endocrine system?
It is the collection of glands that produce hormones that regulate numerous physiological functions of the body such as growth, metabolism, sexual functions etc.
What are hormones?
-made in glands or cells
-activates physiological response
-distant or local target tissue receptors
-cell to cell communication molecules
Hormonal regulation
-Reproduction, fertility, sexual function
-response to environmental situations
-Growth and development
-Maintenance of normal homeostasis
Amine hormones
-derived from the amino acid tyrosine
-include catecholamines (dopamine) and thyroid hormones
-stored until secreted
*Receptor locations
Surface(dopamine)/ Intracellular (nuclear; T3 & T4)
Endocrine drigs
-Hormones are pharmacologically classified as drugs
-can be natural, semi-synthetic or synthetic compounds
-indications:
A. Replacement therapy
B. Treatment for certain disorders
C. Diagnosticpurposes
Antithyroid drugs
Used to lower the functional capacity of the hyperactivethyroid gland and have the following directions of action:
1. Inhibit thyroid-stimulating hormone secretion by anterior pituitary: Iodine
Diiodthyrosine
2. Inhibit thyroid hormones synthesis in the thyroid gland:
Mercazolyl
Propylthiouracil
3. Inhibit iodine trapping by the thyroid gland: Potassium perchlorate
4. Destroy thyroid tissue:
Radioactive iodine
Mineralocorticoid drugs
Desoxycorticosterone,acetate, desoxycorticosterone trimethylacetate, flourhydrocortisone acetate.
Antagonists of mineralocorticoids
Inhibitors of synthesis -metyrapone
Tissue receptors blockers- spironolactone
Hormonal contraceptives
Oral- combinations of estrogens and progestins:
-monophasic combination tablets -rigevidon, minisiston, marvelon
Biphasic combination tablets- anteovin
Triphasic combination tablets-triquilar, trisiston, tri-regol
Parenteral contraceptives- medroxyprogesterone acetate [depo-provera]
Implantable contraceptives- levonorgestrel [Norplant system]
