Effect of substrate concentration on enezyme activity Flashcards
What is the substrate in this experiment?
-thing being broken down
-hydrogen peroxide
What is the enzyme in this experiment?
-thing breaking down
-catalase
Where is catalase found?
in raw potatoes
What is the IV?
hydrogen peroxide/substrate concentration
What is the DV?
time taken from disc hitting surface to floating up again
What are some control variables?
-volume of hydrogen peroxide
-size and type of potato
-paper disc size
-volume of distilled water in paste
Describe the concentrations and volume of hydrogen peroxide
-total volume of 10cm3
-concentrations of 2vol, 4 vol, 6vol, 8vol and 10 vol
-eg 2vol: 2cm3 hydrogen peroxide and 8cm3 distilled water
Describe the procedure
-grind 3cm piece of potato with 5cm3 distilled water to make enzyme paste
-place 10cm3 of hydrogen peroxide solution into test tube
-use forceps to dip filter paper disk into enzyme suspension, tap off excess
-drop paper disc into hydrogen peroxide solution and measure time from striking surface to floating back up
-remove disco and repeat for next concentration
How does this procedure link to the question?
-the catalase enzyme speeds up breakdown of hydrogen peroxide substrate
-time how long it takes for enzymic disc to resurface, meaning reaction is over
What kind of results would you expect to see?
as substrate concentration increases, reaction time increases
